Request 35S::Ac Seeds

In order to mobilize the Ac transposon present in WiscDsLox lines one must cross the WiscDsLox line to an Arabidopsis plant that expresses the Ac transposase gene.  Nina Fedoroff’s lab has generated 35S::Ac lines in the Columbia ecotype that can be used for this type of experiment.  Dr. Fedoroff has kindly provided us with seeds from these Ac lines and has given us permission to distribute them to the community.

Please click here to request seed from the Columbia 35S::Ac lines.

A complete description of these lines can be found in the following publication:

"Resources for targeted insertional and deletional mutagenesis in Arabidopsis." Plant Mol Biol. 2003 Sep;53(1-2):133-50. Zhang S, Raina S, Li H, Li J, Dec E, Ma H, Huang H, Fedoroff NV.

It should be noted that this transgenic line has a visible marker gene on the 35S::Ac T-DNA that makes the plants grow slowly, have lots of trichomes, and look purple.  Although this makes it somewhat easy to identify plants with the Ac construct, it does make the plants grow more slowly.  We have also observed that these phenotypes have variable penetrance.
