              Checking for Nuclei Fibers (Yo-Yo Staining)

 1.   Prepare slides for fiber analysis (as in BAC Fiber-FISH or Nuclear Fiber-FISH protocols).

2.    Dilute Yo-Yo (Molecular Probes) in PBS following manufacturers directions.

3.    Add 100ml of the Yo-Yo dilution to the slide and add a cover slip.  Store in a dark place at room temp for 10-20 min.

4.    Wash 3 times in PBS (5 min each).

5.    Short dry.  Add anti-fade (Vectashield) and cover glass.

6.    View slides.
